Description

Esomeprazole works by binding irreversibly to the H+/K+ ATPase in the proton pump. Because the proton pump is the final pathway for secretion of hydrochloric acid by the parietal cells in the stomach, its inhibition dramatically decreases the secretion of hydrochloric acid into the stomach and alters gastric pH .

Usage And Safety

Dosage

Adults : GERD : For Adults : 20mg or 40mg once daily for 4 to 8 weeks. 12 to 17 years -- 20mg or 40mg once dailly for upto 8 weeks. Gastric Ulcers : 20mg or 40mg once daily for upto 6 months . Erosive oesophagitis : 40mg once daily for 4-8 wk . Prevention of relapse , 20mg once daily . Children : Not recommended . OR As directed by your physician .

Side Effects

Headache , GI upset , Skin reactions , Dry mouth. Rarely angioedema . Rarely angiooedema , anaphylaxis , fundic gland polyps.

Drug Interactions

Phenytoin , warfarin , clopidogrel , ketoconazole , itraconazole , voriconazole , diazepam , citalopram , imipramine , clomipramine , CYP3A4 inhibitors or inducers , tacrolimus , digoxin , erlitinib , methotraxate

Indication

Treatment of Heartburn , GERD , Gastritis & Ulcer

When not to Use

It is contraindicated in patients with known hypersensitivity to proton pump inhibitors (PPIs). Hypersensitivity reactions e.g. angioedema and anaphylaxis, bronchospasm, acute interstitial nephritis and urticaria have been reported with esomeprazole use.

Precautions

Precaution

Daily long-term use (e.g. longer than 3 years) may lead to malabsorption or a deficiency of cyanocobalamin.
